A Golgi-targeted off-on fluorescent probe for visualizing pH change

Abstract

As a weakly acidic organelle, the Golgi apparatus plays an important role in cell proliferation, enzyme activity regulation and ions transportation. When the pH of the Golgi apparatus changes, it will affect the normal function of the Golgi apparatus. Herein, a coumarin-based Golgi targeted fluorescent probe (Gol-Cou) is proposed, which shows significant pH dependent fluorescence at 447 nm and a linear relationship in the pH 5.7-7.1 range. The pKa value of Gol-Cou was measured at 6.39. Besides, we successfully applied this probe to visualize the change of Golgi pH during different conditions in HeLa cells.
